Methods: In order to induce bacterial corneal inflammation in rabbit eyes, an intra-corneal injection of Staphylococcus epidermidis was done. The applied bacterial dose was 50 mL of 1 3 104 cfu/mL. Results: The characteristic corneal change was welldefined ulcer formation located in a limited area and relatively elevated. The area surrounding the infected part of the cornea was rather transparent. Using these infected eyes as models, the intraocular drug dynamics of 0.3% Ofloxacin were examined. The model was successfully induced in 78.6% of the rabbits. The drug concentration in the aqueous humor was 2 times higher in the infectious eyes than in the control eyes. Conclusion: The model might be useful for drug dynamic studies in infected eyes. Regulation of Gene Expression in Cultured Rabbit Corneal Stromal Cells With Recombinant Adenovirus Using the Cre/loxP System Purpose: To control exogenous gene expression is an important norm in human gene therapy. We attempted to control lac Z gene expression in a rabbit corneal stromal cell (RCS) culture system with recombinant adenovirus (Ad) and the Cre/loxP system. Materials and Methods: The RCS was transfected with an Ad which expressed the lac Z gene by excisional deletion of stuffer DNA, Ax CA LNLZ. Four days after the transfection, the cells were also transfected with a Cre-producing Ad, Ax CA NCre. Result: Few cells (0.8%) of the RCS transfected with Ax CA LNLZ expressed the lac Z gene. After the transfection of Ax CA NCre, all cells strongly expressed the lac Z gene. This result showed that recombinant adenovirus using the Cre/loxP system was capable of controlling exogenous gene expression in cultured rabbit corneal stromal cells. Conclusion: We suggest that recombinant adenovirus using the Cre/loxP system is useful for studying ophthalmic gene therapy. Corneal Wound Healing and Immunohistological Features of Extracellular Matrix Following Penetrating Keratoplasty in Rabbits Purpose: To analyze corneal wound healing after penetrating keratoplasty (PKP). Materials and Methods: We performed PKP on 20 white rabbit eyes and applied immunohistochemical techniques. The distribution of type I, III and IV collagens, large proteoglycan, chondroitin 6-sulfate, chondroitin 4-sulfate, and vimentin was determined at postoperative intervals of 3 days, 1 week, 2 weeks, 1 month and 3 months. Results: By day 3, staining for type IV collagen was observed along the host-graft junction. By day 7, staining for type III collagen, large proteoglycan and chondroitin 6-sulfate had increased in the repair region, and then diminished with increasing postoperative time. Epithelial wound healing required more than one month, whereas the remodeling of Descemet’s membrane did not terminate at 3 months after PKP. Conclusion: The data from this study suggest that type III collagen, large proteoglycan and chondroitin 6-sulfate play a crucial role in the corneal wound healing after PKP.
